Chen Y, Li P, Xue M et al. (2026). Moringa oleifera Leaf Extract Alleviates AFB1-Induced Hepatotoxicity and Oxidative Stress Through the PPARγ/Nrf2 Signaling Pathway.. Foods (Basel, Switzerland).

This study investigated whether Moringa oleifera leaf extract (MOLE) could protect against liver damage caused by aflatoxin B1 (AFB1), a dangerous toxin commonly found in contaminated crops like peanuts and corn. AFB1 is a potent carcinogen that poses serious threats to food safety and human health, with limited treatment options currently available. The researchers used both mouse liver tissue and isolated liver cells (hepatocytes) to test MOLE's protective effects. They found that MOLE significantly reduced liver damage markers - specifically the enzymes alanine aminotransferase (ALT) and aspartate aminotransferase (AST), which are released when liver cells are damaged. The extract also dramatically reduced harmful reactive oxygen species (ROS) and malondialdehyde (MDA), while boosting the body's natural antioxidant defenses including total antioxidant capacity and glutathione levels. Using advanced computational analysis, the researchers identified 50 bioactive compounds in MOLE and discovered that key compounds like naringenin, quercetin, and luteolin work by activating specific cellular pathways. The mechanism involves MOLE binding to a protein called PPARγ, which then activates the Nrf2 pathway - a crucial cellular defense system that helps cells cope with oxidative stress. This research is significant because it provides the first detailed explanation of how moringa leaf extract protects against aflatoxin poisoning, potentially offering a natural approach to preventing liver damage from contaminated foods.
